Fast temporal detection of intracellular hydrogen peroxide by HyPer

Abstract

|
HyPer is the genetically encoded biosensor of intracellularhydrogen peroxide (H2O2), the most stable of the reactiveoxygen species (ROS) generated by living cells. HyPer has ahigh sensitivity and specificity for detecting intracellularH2O2 by confocal laser microscopy. However, it was notknown whether high speed ratiometric imaging of H2O2 byHyPer is possible. We thus investigated the sensitivity ofHyPer in detecting changes to the intracellular H2O2 levels inHEK293 and PC12 cells using a microfluorometer imagingsystem. Increase in the HyPer ratio were clearly evident onstimulations of more than 100 ¥ìM H2O2 and fast changes inthe HyPer ratio were observed on ratiometric fluorescentimages after H2O2 treatment. These results suggest thatHyPer is a potent biosensor of the fast temporal productionof intracellular H2O2.
